I know this question has been asked before but I believe my issue is a bit different. I did a set of 6 minipreps from about 1.5mls of culture using the PureLink from Fisher. The nanodrop gave me from 50 to 100ng/ul. The 260/280 was decent, 1.95 to 2.05. The 260/230 was very wacky though. That was ridiculously high, as much as 1000.
I did my diagnostic cuts and ran it on a 1% agarose gel. I cut about 500ng of plasmid. The positive clone should be big, around 17Kb. The weird thing is, one miniprep gave me a very nice clean band. It was negative, but it looked as it should. the other 5 bands do not even show up. There is just a very faint streak. Where is my plasmid? I can understand possible issues if none of them showed up, but that one did and looked great. Also, if anyone has any idea what it means to get a 260/230 ratio of 1000 I would love to know. I know it can be contaminating RNA or DNA but that one plasmid was clean.
