Our last WB membrane has shown an unusual pattern over the bands. I think that they could be scratches but these have appeared in two unrelated membranes (they are from different gels and they have been washed/incubated in different trays) and we have handled them with care.
Any idea? Thank you in advance.
It seems like there might be something off with the image or scanner. I'm not sure if it's related to your membrane, but can you see anything unusual with your eyes directly? It really looks odd.
Also, did you prepare fresh PBST for the washes, or check the water you used? That might help troubleshoot.
The machine works, that should not be an issue. We usually use 10x TBST and dilute it before the experiments. We will try to prepare fresh tbst
