I have uploaded the image of the DNA sample under Atomic Force Microscopy(AFM)
I don't know why my DNA sample shows so many unspecific things.
I used this protocol:
Adsorption of DNA to Mica, Silylated Mica, and Minerals: Characterization by Atomic Force Microscopy,
the buffer solution for DNA was 10mM MgCl2 and 40mM HEPES with pH 7.6.
I filtered all my buffer.
I used Dimension AFM Model DI-3100.
I don't know how to tackle this issue.
Hi Kim, the smaller dots may be due to the contaminants in the DNA sample itself. You may check it by imaging the same mica substrate with buffer alone and not DNA. If the particles don't exist in the image, you would know that they are in DNA sample. Otherwise you would know that they are in buffer solution
Are you sure that the DNA sample is not contaminated? What is the source of your sample?
. I used PCR purification kit to purify my DNA sample.
I did not cleave my mica. I guess that was the problem..
Kim, you are right. Cleaving mica is the most important step for AFM imaging. That should solve the problem.
