Can you explain to us what you're trying to accomplish? It almost seems as if you're reading some articles and asking a question here each time you come to a concept or a phrase that you don't understand. We might be able to help more then rather than dealing with one concept at a time.
Since you have not said what sugars are in your sample and not supplied an example of an injection of a blank, it is very hard to help you.
Personally, I prefer HPAEC for carbohydrates. The sensitivity of amperometry is great, no derivatization is needed and resolution is fantastic. However, they are not the greatest for sugar alcohols. See http://www.dionex.com/en-us/webdocs/114769-BR-HPAE-PAD-Carbohydrate-Analysis-BR700910_E.pdf. But if you are using an amine column I suggest that you follow the manufacturers suggestions. Shodex suggests that the best results are obtained when the eluant is neutral so that the amine groups of the packing material provide a slightly alkaline environment, best for separation.
