Currently, I'm working with the RAW 264.7 cell line. We used to use Trypsin-EDTA to remove cells from the surface of other cell lines, however, ATCC recommended using a cell scraper instead of Trypsin-EDTA or other dissociating agents to detach cells of this type.
Make some suggestions on this issue.

RAW 264.7 cells are semi-adherent, which means some cells grow in suspension, some loosely attach to the surface and others flatten out and attach to the flask.
Treatment with trypsin, however, can impact the expression of proteins on the cell surface and may compromise cell health. To avoid such damage and to improve the quality of harvested cells, dissociation using trypsin should be avoided. Instead, a cell scraper is recommended.
