I am growing SW620 cells in L-15 media supplemented with 10% FBS and pen strep. However 1ml of a fully confluent plate subcultured takes over a week to become fully confluent. Any reason why this is so slow? The population doubling time is supposed to be 26 hours but this does not appear to be the case for my cells.

The complete growth medium used to expand SW620 cells is L-15 + 10 % (v/v) FBS.Complete growth medium (L-15 + 10 % (v/v) FBS) should be pre-warmed before use by placing into a water bath set at 35 oC to 37 oC for 15 min to 30 min . After 30 min, the complete growth medium (L-15 + 10 % (v/v) FBS) should be moved to room temperature until used. Also, check if the cells need any coated surface. For instance, bone cells need collagen coating on the culture plates !
