We have been working on PDAC for 2-3 years now but since a couple of months, my mice do not develop PDAC anymore.
This is the protocol I follow:
- 2 month old mice ElaCreER / LSL-KrasG12D from mixed background (C57Bl6 and CD1) are injected 5x/week with
--> IP 100µl Tamoxifen (30mg/ml in corn oil)
--> SubCutaneous 100µl 4-OH-Tamoxifen (0.3mg/ml in corn oil)
- 1 week rest
- 1 week Caerulein injections: 6x/day on Monday, Wednesday and Friday for acute inflammation (125µg/kg of mouse)
- 1 week Caerulein injections: 1x/day every day to maintain inflammation
- sacrifice the next Monday
We used to obtain nice PDAC, but now we only get pancreatitis and some spots of acinar-to-ductal metaplasia. The KRasG12D mice lose quite some weight during the first caerulein-week, but then regain it slowly on the second caerulein-week.
Does someone have experienced the same problem?
