The cells were growing slower than normal but they were still growing and forming colonies after transfection. However after I added 5% Pen/Strep the cells around the edges start to detach and 'wriggle' in the media. After about 3 hours some of the cells are just lost from the colonies leaving 'holes' in them. Is hESC always sensitive to antibiotics?

5% Pen/Strep concentration is quite high. Usually we add 1% Pen/Strep into the growth medium for mammalian cells.

Hi Dee, stem cells are very sensitive to any change in conditions.
Why did you add 5% Pen/Strep? Was there a bacterial infection?
If there was a bacterial infection then killing the bacteria would release LPS (endotoxin) that definitely will affect stem cells.
