I am using two different systems for my RNAi screen and both of them seem to have 2 origins of replication and I am not clear what the reason is? One of them has a f1 and pUC ori and the other has pUC and SV40 ori? Is there any specific reason for this?

In your case, pUC-ori serves for replication in E.coli, Some vectors (often used as "shuttles" between two organisms) are designed to replicate in several types of host cells. Thus, SV40 origin of replication allows autonomous (as an episome) replication in mammalian cells expressing the SV40 large T-antigen, such as COS-7 cells. f1 stays for filamentous phages. It initiates a "rolling circle" replication in phage-infected bacteria and is useful if there is a need to produce vector as a single-stranded DNA

Hi Andriy but in case plasmid pGP1‐2 and pT7‐1 origin of replications are of p15A and ColE1. what is the reason?
