During Islets differentiation process, most of Islets get settled down into non adherence culture plates. So that i didnt get mature Islets because its staring to de-differentiation. Ideally i want floated Islets through out Islet differentiation Process.
If you mean that the islets are becoming attached to the culture plate, then there are a couple of things you can do. First, make sure that you are using bacteriological grade plates rather than tissue culture treated plates. Alternatively you can add a layer of gelatin or agarose to the bottom of the plate to keep the cells from contacting the plate and to prevent them from forming attachments.
Thanx,robert for reply me.....but I have tried also islet differentiation into glass petriplate still I got problem of islet settled down . Now, I wil try gelatin or agarose coated plates...

Glass is also highly charged electrostatically which will encourage cells to adhere. Anything that you can do to dispel the electrostatic charge on plastic tissue culture plates or glass plates will help to reduce cell adhesion. This includes pretreating the plate with FCS, PEG, or BSA for several minutes followed by washing with your culture medium, if your cell system will tolerate this.
