most of the author quotes that lutein and its isomers are best identified in and around 420 -450 nm range in the sample. but in my case I have been getting peaks at around 300-350 nm range..I dont know what this peak resemble....but has to confirm what it is......nd also can anybody suggest me or provide me a standard graph for lutein.....
Since you are getting absorption bands around 300-350 nm range, it shows that your sample contains some UV absorbing components. Your expected range of 420-450 lies in the visible region. Samples absorb in UV range because of the presence of some Chromophores. If the samples contain multiple chromophores, the absorption maxima gets extended to longer wavelengths into visible range and then it becomes a colourimetric estimation. So the absorption range ( whether 300-350nm OR 420-450nm) will depend upon the chromophores present in the sample and their number ( whether a single chromophore or multichromophore).
It is important not so much the quantity of chromophores, but their interaction (Conjugated pi-bond system chromophores). Chromophores no interact in your compound!
