Hello everyone,

I treated my cells with different concentrations of a desired drug and measured the concentration of glucose in the supernatants using a Varioskan microplate reader. To do the measurement, I first diluted the samples (1:2), then I piepetted 5 ul of the sample into 95ul of the reagent. The glucose concentration in my control group is 2 mg/ml. However, the ODs that I get from the machine shows that the glucose concentration in my control is more than 2mg/ml (like, 3.99 or 2,98 mg/ml). What could the problem be? Thank you.
