For latex nanoparticles, conjugation to proteins can be done either covalently or non-covalently.
The covalent method is usually long (days) and complex (NHS, cabodiimides, etc), but the non-covalent is done through absoption and can be quick (1 or 2 hours).
This being the case, why is the covalent method the most popular?
Physisorbed materials exist in equilibrium. i.e., if the bulk conditions change, the material can desorb. Chemisorbed materials (i.e., covalently bonded) won't.
Thank you. That makes sense for a range of applications, however, I was wondering if the physical method could be more time and labour efficient under more controlled condtions.
Also, the efficiency of conjugation and quality of conjugate could be different, bu im not finding good comparative studies about this topic.
Also also, Could it be that this method is just less known and therfore, less used?
