I have purified a protein with a C-terminal NLS and labeled this protein with Cy5. I tried to deliver this labeled protein into the nucleus of HeLa cells via electroporation. After electroporation, I allowed the cells to adhere to the plate for 10 h before analyzing the signals using an LSM880 microscope.
Results showed that my protein was trapped in endosomes (fluorescent signals in drops in the cytoplasm), and no detectable signals in the nucleus.
I have tried various electroporation conditions, but all tests have failed to find nuclear localization. Have you encountered similar problems? If so, I would appreciate it if you could share any strategies that worked for you,
