I repeatedly get LDH results where the controls are always lower than the treated samples? (72hrs incubation)
eg: cont.: 0.08,
S1: 1.6
S2:1.2
S3:0.6
I guess you mean LDH for cell damage. Maybe the treated samples had more cell damage than controls. Hope this is the question you asked.
As Mr. Teng mentioned, what do you mean by LDH?
Lactate dehydrogenase and cell injury? then I am agree Mr. Teng, since LDH used for long time to measure cell damage soif your intervention induce any damage then it is logical to measure higher LDH in compare to your Ctr.

I am confirming the MTT Cytotoxicity results by measuring Lactate Dehydrogenase activity after cell damage.
The problem that I have no explanation to the low results for the LDH in the control.
eg. (MTT 1.CONTROL=.0.8 2. DRUG CONC.1= 0.6 3.DRUG CONC.2=0.4 4.DRUG CONC.3=0.2.)
(LDH 1.CONTROL=.04 2. DRUG CONC.1= 0.7 3.DRUG CONC.2=0.5 4.DRUG CONC.3=0.3.).
Basically, your drug should be able to inhibit the damage or induce damage?
Please note that MTT results could be influence by any interventions which can have an effect on cells mitochondria, for example if you have Ctr in which your cells are not active due to absence of FSC or etc, then your results will be low. In addition, time points are important. Dud you checked your cells under the microscope to see if there is any differences in the phenotype?
