The very same amount of GAGs will show higher percentage in decellularized samples since the native tissues have more things in them, i.e. they will be more diluted. Maybe determine the amount of GAGs in something else than percentage.

The amount of ECM components such as GAGs or collagen will be higher in decellularized tissues than native tissues because of loss of other proteins. Attached is our team's explanation about this point (published as supplementary data in provided link).

 Dear Janani, both the above are correct. We also observe the same with pig liver. If you want to  get meaningful results you should carry out the GAG and collagen quantification in samples which are fully swollen in PBS or other isotonic solution. In that way the "holes" you make when decellularising are filled with water, so the tissue mass remains essentially unaltered before and after cell removal. We have described this method in our paper.

 Thank you Kamal El-Din Hussein for the reasons provided here for the GAG content more in the decellularized samples. This is a good reasoning for more extracellular matrix content in decellularized tissues.

  Thank you Arti Ahluwalia. I am also decellularizing porcine liver for my work. I will try the GAG and collagen estimation from swollen samples. I have found, there is no collagen retained in my decellularized samples (estimation done in swollen samples). I used Trition X-100 for decellularization. Kindly help me in this.

How did you measure collagen? We used BCA and Sirius red. We found the livers are almost 100% collagen (dry weight). I can send you some protocols if you like.

Thank you very much Arti. We measure collagen based on Sirius Red Dye method. Native liver samples are showing collagen content, but there is no collagen in decellularized liver.  I am thankful if I have some protocols. I will follow it.

A relative increase of insoluble scaffold components is expected in decellularized samples with respect to native tissues. However in the case of liver the rate of GAG increase even if consistent with the recent literature, it does not exclude the risk of an overestimation of the residual GAGs. In fact in such lit reports the rate of GAG conc increase exceeds by far that otherwise expected for collagen which is the main component of residual scaffolds. Conversely in many other tissues the decellularization procedure results in the loss of GAGs. Moreover, unknown analytical factors possibly related to the still uncertain amount of residual detergent bound to the resulting scaffold points to a more likely attitude of caution. In addition the more reliable method of collagen estimation remains for sure the determination of total hydroxyproline conc.
