Hi everyone,

I’m using Cleaved-Caspase 8 Antibody (YA792) (HY-P80624 from MedChemExpress) in MCF7 cells to perform a Western blot after heat stress treatment. However, I’m running into a few issues:

  • The band for cleaved Caspase-8 is very weak, even after trying both weak and strong ECL reagents (with no significant difference).
  • The band appears around ~35 kDa, but I’m unsure if this is the correct size. I expected the full-length Caspase-8 band at ~55 kDa and the cleaved fragment around 43 kDa, but my results don’t quite match this pattern.
  • The overall background is low, but the specific band is faint and not in the expected position.

Does anyone know if certain sample preparation steps are required for using cleaved Caspase-8? Any suggestions or shared experience would be very helpful, thanks!
