I am planning to do an MTT assay for the cytotoxicity evaluation of several plant extracts on A431 cell line (human epidermoid carcinoma). For this I need to use a positive standard. In the past I have used staurosporine, but it is very expensive (I am PhD Student :) ). Are there other substances that I can use that have proven to have a cytotoxic effect on this cell line? A431 cancer cell line (skin carcinoma, human)
Cell cytotoxicity was measured by experimenting MTT (3-(4,5-dimethylthiazol-2-yl)-diphenyl tetrazolium bromide) assay (Mosmann 1983).
You can try using it as a positive control H2O2 (20 μL) was used as positive control and also to induce free radical damage in the A431 cells,
There is no need to maintain a positive control always.
The simple standards for MTT assay would be A431 cell untreated with plant extracts (Negative control) ( Cells left untreated + MTT + solubilizing buffer)
As a blank, maintain medium with no cells ( NO cell + MTT + Solubilizing buffer)
There is always a way when budget is an issue.

You can perform your experiment using untreated cells as control where cell viability is considered to be 100%. Therefore, cell viability of treated cells is expressed as percentage of untreated cells.
However, if possible, you can used cisplatin as positive control.
