I have a DNASep cartridge (DNA separation column uses alkylated non-porous polystyrene-divinylbenzene (PS-DVB) copolymer microspheres for high performance nucleic acid separations). I want to know which kind of solvent can dissolve or damage this column?
especially acetone and n-butanol.

Dear Leila Rastegar, St/DVB resins can only swell in some solvents. If you 'digest' this resin with strong chemicals you will loose your DNA as well. Since it is a non-porous adsorber, this means that you DNA is only surface adsorbed. Desorption is the simplest way, just do simple search on 'desorption from solid support'. My Regards

thanks for your guidance. You know, I have a DNASep column, as I described that. I set a method for mRNA analysis. buffer for elution is a gradient of TEAA 0.1M & 25% ACN. column oven is 75 C. after method validation, I realized that the result is not reproducible, and I cannot get the mRNA peak. the first possibility was damage on the column. but where was the source of damage? I found that we filter TEAA 0.1M & 25% ACN buffer with MCE (mixed cellulose ester) filter. this filter is not recommended for acetonitrile. I do not know what happen when I use this filter. another parameter that can affect on column and result is sample. maybe there is some compounds in sample (e.g., glycerol) that damage on column. but I tried to control concentration all of them in the sample.
