Which service would you use to get 500-600bp of synthetic DNA?
Low price, fast delivery and high quality are important for me. I plan to order 500-600bp which I plan to use as a template for PCR and not direct cloning. I was considering IDT gBlocks or GeneArt Strings. They are cheap and fast to deliver. My only concern is that they are only bulk sequenced. Therefore, it is expected that only 80-90% of the DNA I get is the exact sequence I ordered. Does anyone have experience with them?

Hi! I ordered gene strings once and it worked perfectly OK! It's cheap, it was fast and very easy to design with the online tools they developed on the website. No issues with the sequence, it was exactly as ordered. I can definitely recommend it!
GenScript does gene synthesis and has low prices, you will get it automatically in a pUC-based vector so you could always PCR or subclone from that. Free codon optimization. For 500-600bp it would be around 150-200$ I believe.
You can synthesize the fragment using assembly PCR. For this order fragments with 100 - 150 bps manner that each fragment overlapped with next and previous fragments in 20 nucleotide. Do PCR the fragment together.
Assembly would be problematic due to some repetitive sequences. I think that once I will design the final DNA sequence, I will get a price quote from IDT, Geneart and Genscript.
