CD14 positive monocytes were needed for transmigration experiments, thus no CD14 positive selection is possible due to activation by this method.
Stemcell has a kit (EasySep™ Human Monocyte Enrichment Kit) with reported purities of 83-95%, starting with frozen PBMCs. They also have another kit which keeps the CD16+ monocytes also.

I agree with Nuruddeen. We use Easy-step kit negative selection in my lab on buffy coat. We have more than 90% of purity without activation. It's a realy good product.
