I'm designing a cytotoxicity assay to measure the effect of treatment on proliferation. My specific objective is to compare the number of cells with 1 and 2 divisions in treated cultures to untreated controls. I'm using Tk6 cells, a lymphoblastoid cell line.

I am considering a dye-dilution method, with the quantification algorithms built into FlowJo.

The cells would be:

  • Stained with the dye then resuspended in growth media
  • Treated for up to 24 hours untreated cells are expected to undergo one or 2 cell divisions during this period, the cells are not synchronized so the population will be mixed.
  • Analyzed by flow, using FlowJo's proliferation model to estimate the number of cells in each division
  • I'm considering the following dyes, I was hoping someone could recommend which one might perform the best for my intended purpose or another dye that might outperform the ones I have listed.
