I would like to prepare nano-formulation to target liver cells, but I am not sure which would be most appropriate to deliver the drug to desired site.
Most particles with the size small enough to escape from being trapped in the lungs (less than approx 7 microm) will stop in the liver (and also some in the spleen) but all are taken by the fixed macrophages in RES. If you want to target other cells in the liver, the first thing is to avoid this, which can be achieved by generating a steric PEO barrier around the particles like long circulation stealth liposomes. Once you have achieve this then you can attach a site specific antibody or its fragment or other binding protein for targeting to the special cells but this will be a big challenge.
Nanoparticles clear to the liver when they are deliver systemically. To target liver cells specifically, it is important to focus on the particular targeting agent, such as an antibody or protein, in addition to the nanoparticle.
