I need to perform qPCR validation of miRNA. Which methods are suitable for RNA extraction. Does RNA from Himedia RNA extraction kit be appropriate or I should use some other method?
Most modern RNA column kits will quantitatively retain the small RNA fraction, but some don't, so check the protocol and the manufacturer's notes before purchasing. I use Zymo kits and they work great. You should avoid bead-based methods (RNAClean XP) and alcohol precipitation steps, because these generally won't retain your small RNA.
All the suggestions from peers are wonderful. Any product from Qiagen for miRNA is best. Starting with a miRNA extraction kit is good. Well, I have tried one more way.
You can isolate total RNA using TriZol and then use Qiagen miScript RT kit II for cDNA synthesis. The kit basically converts all miRNA present in particular RNA samples into miRNA-specific cDNA. That cDNA can be used directly with SYBr based qRT-PCR. So, here if you get different miRNA primers of your interest, you can analyze them with that miRNA-specific cDNA with SYBr based qRT-PCR.
NOTE: I. Here using that Qiagen cDNA kit (Qiagen miScript RT kit II) only, you can go for SYBr. Since many including me found that the Thermo miRNA advanced cDNA kit generally does not work for SYBr, so you have to get the Taqman probe.
II. Prefer pre-made miRNA primers from Qiagen, which comes in LNA tech-based and ready to use.
