Which method do you use to determine the contamination by mycoplasma for HUVECs?

I have to do a mycoplasma assay for the HUVECs (Human Umbilical Vein Endothelial Cells) and I used the MycoAlert Mycoplasma Detenction Kit (Lonza). I have to do a cell culture for a week without antibiotics but the usual cultural medium that I use (ECGM, Endothelial Cell Growth Medium) contains already antibiotics. I tried one time with DMEM and one with F-12 K Nut Mix, but in any case all cells died before the test. Could anyone help me?

HUVEC always need reach media...they eat a lot! You cannot use DMEM

(I think LONZA sells different medium for endothelial cells without antibiotics)

...by the way, I usually check mycoplasma contamination with a simple PCR (way cheaper than commercial kits)
