I make a concentration of 50 ng/mL VEGF in endothelial cell growth medium. I add this medium to my microfluidic device to stimulate HUVECs for angiogenesis.
I was wondering how long I can use this media? Does the functionality of VEGF reduces over time?

VEGF experiences loss of activity in vitro possibly due to factors present in the serum. Also, VEGF has been demonstrated to degrade in isolation, with a half-life of ~96 min in acellular conditions at 37°C.

I would suggest you add 50 ng/ml VEGF fresh each time to the endothelial cell growth medium whenever you have to add this medium to stimulate HUVECs.
