I want to analyse animal blood for identification and quantification of organic pesticides(organochlorines, pyrethroids and organophosphates). However I've seen many collection and storage protocols, some use plasma, other serum and others whole blood with some anticoagulant such as EDTA.
I've already have several frozen samples of whole blood stored with EDTA. I'm wondering if this is the recommended protocol for this kind of analysis.
If you have a validated protocol for assay, just follow it! However, if you use whole blood, you measure total pesticide in plasma and blood cells. Between serum and plasma, I prefer serum, because it has less protein and no anticoagulant. As a general rule, the more purified sample medium, the more accurate assay. So you may face less interference in the assay when you use serum. On the other hand, it is possible that some of your target materials bound to coagulant protein or RBC surface, so they will committed from measurement. Check this hypothesis!
