Hi to everyone.
I'm planning to set up angiogenesis experiments with HUVECs. Usually, I grow them with EGM2 bulletkit (Lonza) supplemented with 2% FBS and other factors included in the kit.
Now I want to test the effect of a compound on tube formation, as well as on transmigration, so I was wondering if I have to use EGM basal without growth factors and serum. I hope someone can help me.

According to our experiences, you have to use the full medium for tube formation. Otherwise, HUVEC can not form tube without serum and growth factors.

Hi Thomas, would you advice serum starvation also for a 96-well plate proliferation assay? if i plate the cells in the treatment conditions, i see clear differences (not always what I wish to see). if i plate cells with serum, and change to treatment conditions (with no or reduced serum, but other angiogenic factors, VS. control with serum) then I don't see strong differences. Any comment on that?
Also: when you say "only with bFGF or VEGF"... isn't proper HUVEC medium supposed to always contain also heparin, EGF, hydrocortisone, and FGF?
