Hi everybody, I was wondering which is the best way (protocol) to dissolve 4-Hydroxytamoxifen.
I would also like to know how can I store the reagent to keep it stable and useful for future experiments.
And finally, for how long is it active at 37C for cell culture. I am planning to run 4 days with 50 and 5 nanoM 4-HTM. Does it sound reasonable?

I usually dissolve it in absolute ethanol. It may be hard to dissolve so you may want to heat up the samples at 55 c degrees for few minutes to ensure a good solubility. If you plan to use it for cell culture, don't forget to filter the solution and make aliquots that are stable (at least few months) if kept at -20 c degrees.

Here is what it is stated:
Soluble in ethanol (20 mg/ml, with heating) and in methanol (10 mg/ml) producing clear faint yellow solutions. Solutions should be stored protected from light at –20 °C. 4-OHT undergoes a cis-trans (E-Z) interconversion process favored by solvents of low dielectric constants when exposed to light and when incubated in culture medium.1,16 This isomerization occurs in all common laboratory solvents, but can be prevented by storage of the compound in tetrahydrofuran containing ~0.025% butylated hydroxytoluene (BHT) at –25 °C in the dark. I have dissolved 4-OHT in ethanol, storaged it in -20, and used it for 24h 37c and it worked fine. Check my paper and the other one. Good luck
