I am working on Stevia rebaudiana. I have gone through few research articles where water, methanol and ethanol have been used for the extraction procedure. Can anyone suggest that which is the best solvent for the extraction of steviol glycosides, especially stevioside and rebaudioside?
You have follow the general protocol of extraction. Starting from non-polar solvent ( n-hexane or petroleum ether ) followed by ethyl acetate then 70% methanol.
Take TLC for three fractions, evaporate the solvents, separate and purify using different available chromatographic methods ( best one is MPLC ). Identify the single compounds using IR, NMR, MS,.....etc

Based on my experience, Ethanol and its binary solutions are the best solvent. Methanol has not GRAS status from US-FDA while Ethanol has GRAS status and is best to use for enhanced extraction of steviol glycosides . I 100% endorse the opinion of respected Prof. Frank T. Edelmann
