I have to order a new product to perform lipofection. So far I used Cellfectin II reagent from Invitrogen but not sure this one is actually the best one as well. I need to transfect insect cells as well as reptilian cells with rather large DNA fragments of 30 Kb to 150 Kb. What are your recommendations?
There's a difference between generic reagents, those for specific cell lines, those for in vivo use, and those for primary cells (e.g. AltoFect: https://altogen.com/product/altofect-transfection-reagent/). There's no one that will be the best for all three categories, and depending on the goals of your research you may need to use different ones for different tasks. Generics are helpful only if reagents designed for the specific cell type are unavailable.
Lipofectamine 2000 is a good old reagent that works really well, and was cited thousands of times over th elast 16 years. it is number one reagent for plasmid transfection, world wide. lipofectamine 3000 is a recent addition to the family, it works better in tough to transfect cells
PEI is a good transfection reagent that works in a number of cell lines including insect. We used PEIpro and had great transfection efficiency in 293T cells.
Lipofectamine or Lipofectamine 2000. I would use Lipofectamine for more robust cell lines and Lipofectamine for less robust ones.

The efficiency depends on the cell line you want to transfect. There are few papers comparing different transfection reagents but in human cell lines. I recommend you try more that one.
