We have got good experience with BD, R&D, DAKO and Biolegend. Abcam antibodies are good but often too expensive comparing to others. Anyway, you should always check whether this specific Ab has been already tested for your purpose, i.e. IHC-paraffin or frozen. The preparation of your tissue is also immensely important: the duration of formalin fixation, purity, embedding technique, etc.

There are many companies that have good anti-mouse AB. I agree with Elena Rosca about Cell Signaling and R&D. Unfortunately some time you have to pick companies that have an AB for what you are looking for regardless of rep because they are the only ones who make it. If you are characterizing common proteins I would stick to these companies, maybe Abcam too. Accurate Chemical has a lot of virology/ID AB's. Vector is good too.

During my research I have worked with Abs from Dako which are great for IHC purposes. Regarding the cell signalling, I have only used their Abs for assessment of signal transduction pathways activation in western blot and they have worked very well.

I have had very good experience with AbD Serotec and Abcam. I specifically used antibodies for IHC and got great results.

I've had better luck with monoclonal Abs than with polyclonals. If you prefer buying from a particular company, you can just search their website. Some companies specialize in IHC reagents and can provide data online, in addition to citing publications that have used their Ab. Some also provide user reviews. Some Abs may be "predicted to work" based on bioinformatics, but have no supporting data. Be careful when buying these, as they usually don't come with a warranty. One company may warranty an Ab for IHC because they have tested it, whilst another company sells the same Ab cheaper with no warranty because they haven't tested it for IHC, but it will work anyway. I addition, not all IHC conditions are the same. Tissue processing (eg, fixation) may be critical, and antigen retrieval may be necessary (especially for IHC-P on FFPE samples).

It is important to compare prices (value). For example, 100ug may cost $200 but 500ug is only $300. When stored properly, Abs keep well past their expiration date, so judge your future needs. Also, the same Ab may be manufactured by one company, and licensed to others. One company may sell 100ug for $200, and another may sell 100ug of exact same Ab for $275. A bit of presearch may save your research.

You're welcome, Sampada. Companies such as Abcam and DAKO have very good technical sheets you can download (eg, "DAKO IHC Staining Methods"). One of my favorite guides is by Rodney Miller, which I have uploaded as an attachment. You will discover that there is a lot published on this matter, and you should do a bit of literature research before you plan your experiments.

Thank you Anna Kovtun and all of you. I am looking at buying secondary anti-rat antibodies HRP and other reagents separately as there is no good detection system kit that is validated on mouse tissues. Biogenex is validated only on human tissues.
