Hi everyone, I will appreciate if you can help me to understand how to extract the spicule from Porifera for the subsequent taxonomic classification.
Which chemical solution I need, which tools, which protocol. In my laboratory there is no expertise in this topic and we are approaching it right now, so please, we need a detailed procedure.

Actually, it is some what long way, but in general following :
1-Collection, drying, grounding, extraction ( either using ultrasonic bath or maceration or sohxlet apparatus.
2-Start with nonpolar solvent ( hexane or petroleum ether, then ethyl acetate followed by methanol finally with 70% methanol.
3- Take TLC for each fraction
4-Separate the single pure compounds using chromatography methods ( MPLC or Column Chroma.)
5- Check the biological activities for each compound, according to your need.
