I'm currently working on production of industrial enzyme via SSF. There is an ongoing argument on fungal inoculation either using plug inoculation or spore suspension inoculation methods. So my question is, which one is better in terms of efficiency and specificity?

There are pros and cons for each method.
Usually a working cell bench of spores is a stable thing, you can easily prepare them and the set a defined OD for starting, they are extremly stable and you have no problem to inoculate a defined number of equal "starters". On the other hand: some fungus change there metabolisms and activities during sporolation, so if you want to raise them just for growth, this will not be a problem, but if you want to make a regular agitated fermentation, there might be a time you need to plan for an iniatial out-of-spore growth and mycel forming before you achieve activity. Usually, this could be solved in a preculture.
If you want to use mycella for inoculation a physical method like adding detergent (Carbopol 934) can reduce the length of the formed mycel and might give you a spore-like, homogenous distribution, I think this can work on agar plates too.

Thank you Mr. Hoger for your suggestion. Maybe the reason that I managed to figured out is that in plug inoculation it provide both spores and mycelia while in spore suspension only spore is present.
What I can hypothesized is that using fungal plug inoculation method provide faster fungal growth in SSF fermentation set since the pre-existing mycelia in the fungal plug inoculum rather than spore suspension method.

Hi mr nizzal, currently i am working on biodegradation for my fyp. Then, which method would u suggest for the inoculum preparation? I've done the spore suspension and it took a long time for me to adjust the hemacytometer(cause i can't find the square) & to count the spore.

That would be depends on the objectives of your research and how the biodegradation is conducted.
