Which DNA extraction or purification steps are influence on yield of DNA? And how to overcome from those steps to have more chance to get higher yield of DNA. This may depend on type of sample and/or some step might take more time, etc. Please share your experience how to solve problem in DNA extraction.

I use DNA extraction kit (column) as a main method and sometime I also extract by manual method too.
Sample type: Anti-coagulant blood (heparin and EDTA), Clotted blood, Animal tissue, Gram positive bact, Gram negative bact, Higher bact, Mycobacterium tuberculosis, and Fungus (yeast and mold).

What kind of protocol do you use when extracting DNA?
Hi, Ana is right. without knowing your DNA extraction method (f.g. CTAB-method, kits) it's difficult to help you. What kind of material you are using (fresh material, dried material, material in ethanol,...)? Plant or animal tissue,...?
The answer is no easy , because:
1- What kind biological sample do you use¨? plant tissue, animal, bacteria or fungus
2- Your material is fresh, dried, dehydrate or another method conservation?
3- What kind DNA extraction method do you use: Kit extraction or manual protocol.
But if you use organic sorvent like chloroform for eliminated proteins you lost yield to DNA too.
