I am planing to do some genome editing with CRISPR/Cas9 in Lactobacillus delbrueckii subsp. bulgaricus (ATCC11842), but I am not sure which CRISPR/Cas9 is suitable for this bacteria strain? Does anybody has experience with this?
Hey, take a look at the attached Link. They used CRISPR for genome editing in lactobacillus reuteri. Basically it works like in any other organism, where you determine your target with the appropriate PAM sequence. The most important thing is to choose the correct replicon and promoter that will work in Lactobacillus delbrueckii subsp. bulgaricus (ATCC11842). What I mean is, that it is not important which CRISPR you choose but the correct and stable expression.
