It would probably make sense to take inoculum from a stand (or multiple stands) of the same tree species with otherwise similar environmental circumstances as the place where you would consider planting (if you are considering planting trees). This would ensure that the AMF effects are relevant to your study. Comparing this to the place where the plantation would take place would be very interesting in my opinion, because it would give you an indication whether the site would benefit from inoculation (if e.g. effects are more positive when seedlings are inoculated with inoculum from a mature stand).

I hope someone else can provide you with the publications of interest, I am not entirely up to date with the state of the art of tree-seedling inoculation research. If you encounter access issues you could of course contact me.

You can trap AM with green amaranth and pepper (Capsicum spp) plant. this can be used to multiply pure AM that could be used for your plantation experiment.

There's actually a product being sold in gardening stores called Alpha Thrive, which is supposed to be liquid inoculum. No idea what species of AMF it uses, though.

to get AM bacteria from the roots of trees will need to find the trees, especially the young roots of the first order with nodules. Plants removed from the substrate or soil, and roots with nodules pack the bags and immediately stored at 40C. In the laboratory the roots washed plain and sterile distilled water, and nodules separate the part of the root. Nodules were surface sterilized with 0.1% HgCl2. Excision of nodules sterile knife ekskudat was seeded on yeast mannitol agar supplemented with Congo red indicator (Shah et al., 1995). After growth for 3-4 days at temp. 280C cultures were transplanted to specific substrates, and single colonies were checked in terms of morphology, whereas the cells of bacteria tested by Gram (Rao, 1999) and then grown in YEM nutrient solution with the addition of brom-thymol-blue (BTB) (Vincent, 1970 ).

You may isolate the AM from the host plant and then the pure culture of the inoculum will be multiplied in any fast growing cereals having fibrous root system for further study of host plant studies. For this you may visit to ivam website for detailed protocols and methodologies.

If you would like to study the effect of the native AMF you can isolate from the rhizoshpere by wet sieving, I roughly remmmber most AMF spores could be captured at 50 micron sieve, most Glomus, Aculospora, etc.,  spores are about 100 micron in dia and only those of Gigaspora are larger. On isolation, inocula can be multiplied on host crops such as finger millet, Eleusine coracana  for further use. Collection sites, that are relatively poor in soil fertility,  away from intensively cultivated areas in my openion are better suited. Alternatively, INVAM at West Virginia University maintain a fairly good collection of AMF, another source is the BEG, International Bank for Glomales at France.

in fact i was not asking how to prepare AM inocula.and the need for a host as it is an obligate symbiosis is clear.however i have got my question answered.thank you all. Manamohan's answer is more relevant to my question however.so was Erik's answer

if you want to study the changes produced by mycorrhizal inoculum land must use the definite place of planting. You can also use in comparative commercial inoculum with the best known species of mycorrhizal fungi.

It is also very important to use plants with roots unlignified to facilitate fungal infection

It has been shown that different species of mycorrhizal fungi can colonize a single plant species, I have yet to experience that certain species are more efficient than others. So I recommend you start your experiment with a known plant species and different species of mycorrhizal fungi, maybe this is a first step.

are you working on a specific area ? or with a large scale ? what do you really want to test ?

depending on that, prefer a fungi' strain from a bank (in vitro) or from field (without soil polution in fact)

i guess it is better to inocculate seeds than "young" roots.

and do not forget about soil community that could slow fungi recruitment (by "myc") or AM development/running if you conduct you experiment outdoor

I remember some of the groups use onion and maize roots for natural inoculum of AMF just by chopping the roots in small pieces and use in the treatments

It's always better to use native isolates as they are ecologically adapted to the habitat,where you intend to carry out your experiment. There are well established isolation techniques that you can get from journals. 

I agree with  Dhruva K Jha . I think you should try , with native mycorrhiza and then with Foranea mycorrhiza .

Better work with native isolated, but I would choose the easiest to get. What you should avoid reinfection is occurring in the place chosen to investigate.
