Yes, we have used this method in the past and it works well for difficult root tissue. However, I would recommend 5% NH4OH for 15-20 mins and repeat as necessary. I think your suggested concentration of 0.5% would be too low.
I believe that the protocol, you should standardize it, testing increasing concentrations of Nh4Oh or H2O2. The color of the roots is due to the content of tannins and different pigments, therefore, each laboratory must always carry out this process of standardization. I hope you have successes
