Hello, I am currently experimenting on the PTH1R and i wish to tag the N-terminus. I will be using sf9 cells and wanted to see if different signal peptides could increase expression (hemagglutinin gp64 etc).
According to uniprot.org PTH1R signal peptide is residues 1-26. So should I truncate this section and continue from 27 onwards?

Yes. If you want to keep the folded extracellular domain of PTH1R intact, you need to keep the original boundary between signal peptide and mature protein. Modelling shows that the new N-terminus is rather tucked in between the extracellular domain and the transmembrane helix bundle, therefore you have to be careful if you add any additional N-terminal domains.
