Hey guys,
I recently synthesized an RNA oligo from IDT which has the same sequence as the mature miRNA that i am interested in, but when i transfected the oligo into the cells i didn't see any effect on its target. Then other people tell me that the miRNA mimics should be a double-strand complementary oligo instead of a single strand that I synthesized from IDT. So how do you guys usually get your miRNA mimics? Also, is the double stand more stable than the single strand?

I work in miRNAs analysis.
The double strand for miRNA mimic is important to coupling in DICER system and then one strand is active to suppress the target gene, like a miRNA.
if a single strand is internalized to cell, it is unstable and it will be degraded. At least that have chemical modification

Thank you, Oscar! But i transfected what i believe is the mature miRNA so it probably has nothing to do with DICER system? I am really confused when people tell me that the mimics should be double-stand. What the double-strand oligo looks like? Let say i am interested in mir-11-5p, is the double-strand sequence mature 11-5p sequence and it's 100% complementary sequence or it's like the stem-loop double-strand which the sequence is not 100% complementary to 11-5p?

Good point of view. But, the mimic is double strand, because this have to mimic (to into) the processes of the natural pathway to mature miRNA in the cell and to use this function cell to get the suppress action of miRNA vs mRNA. If your mimic is single strand, it will be outside of this system and it will be not functional miRNA.
Your mimic sequence should be the stem region of your mimic, in this is included your mature sequence already to into in the system of maturation miRNA.
For more information, look the web site of Qiagen about mimic. I have experience with overexpression of miRNAs and suppression too with this trade mark.
