I'm trying to isolate yeast for further study, can I know where I can isolate it? after isolation, I'm going to grow yeast in fermenter, what is the optimal growing condition for yeast in term of media, rpm, temperature & pH?
Sorry if i may be blunt but i would say you straightforward that the question you have asked are basic question required for foundation of any work and you need to figure out the answer by yourself. The ans are awailable in google, you need to read it by ourself. If you will start depending on answer of other from now than in future you will be always depend on other.

You can use a liquid medium such as malt extract broth the temperature of incubation usually is 25-28°C and pH 5.5. To increase the growth you should maintain the cultures in continuous shaking conditions.You can found yeasts everywhere: on fruits, soils, water, air........
You can isolate wild strains from grape skin. Since significant enumeration occurs while fermentation, you can provide conditions with 1% yeast extract, 1% peptone, 20% glucose. If you already have samples, use YPD agar supplemented with 0.1% chloramphenicol.
