@ Daya Pavitrakar: The point is that influenza virus has negative-sense RNA and replicates in cell nucleus, so rescue of virus from cDNA requires synthesis of viral RNA and at least 4 viral proteins to form ribonucleoprotein complex. As I see plasmid-based reverse genetics of influenza most often employs a bidirectional vector developed by Hoffmann et al. (http://www.ncbi.nlm.nih.gov/pmc/articles/PMC18566/) with promoters and terminators for transcription of both vRNA and mRNA.
