I used sodium nitrite, aluminum chloride, and sodium hydroxide for the total flavonoid assay. I performed the experiment with four different extracts (methanol, aqueous, chloroform, and benzene) of the plant sample. In methanol and aqueous extracts yellow-pinkish color solution is formed but in chloroform and benzene extracts light greenish colored solution is formed and showing a high absorbance value as compared to methanol and aqueous extracts. Is the absorbance of chloroform and benzene extracts is due to high flavonoid content or it would be the absorbance of the reagents used in the experiment.
