I performed TPC and TFC assay for my plant sample in polar and non- polar solvents.

In non-polar solvents (chloroform & benzene ) higher value of absorbance is noted. But antioxidant and antimicrobial activities are not in co-relation with TPC and TFC.

I just want to know that the color of the sample is interfering with the absorbance or not.

I also checked the absorbance of the sample alone (before adding the reagents required for experiment).

Is it relevant to use the following equation to get the actual TPC and TFC of colored sample?

Abs. of sample (with all required reagents) - Abs. of sample alone (before addition of reagents) = abs. used to know the TPC/ TFC of sample

for e.g.

Abs. of chloroform extract with reagents (2.456) - Abs. of sample alone (1.944) = 0.512

Here, I used abs. value 0.512 to calculate the actual TPC.

Is it ok to use this formula or not ?
