Instead of seeding say 10 x 100mm dishes is it possible to just seed one dish with 293t cells and then transfer the transfected cells into a bigger bottle or a new environment?
how much virus do need to produce? I have not tried to trypsinize transfected, virus-producing HEK cells; and I am not sure if I would try because they clearly look stressed. However, they generally produce huge amounts of viral particles making it possible (if not even neccessary) to dilute the supernatant before transduction. No. You are transiently transfecting the 293 cells with 3-4 plasmid vectors . A cell containing all plasmids will produce virus. If you try to outgrow the cells, the daughter cells will begin to lose plasmids and as a result will no longer produce the virus.
