Definitely you need the B27 plus retinoic acid because this and others short chain fatty acids is the more natural way to feed the colonocyte to behave naturally in lab.
I've now received a response from R & D systems' technical service:
"I believe that working in Vitamin A Free conditions is necessary for neural stem cell expansion as limited and controlled retinoic acid production reduce unwanted NSC differentiation. I do not think it is necessary to use Vitamin A Free Media Supplement when culturing human colon organoids. I believe that in the case of those few protocols were vitamin A free material was used with colon organoids it may have been that it was available and therefore used but not strictly necessary. To be sure I am giving the correct answer I am checking with the people that develop the culture media and supplement and I will let you know more soon as possible also about the N2."
