I have been trying to measure mouse plasma/serum IL-12 by multiplex TH1/TH2 cytokine kit (MSD). And the IL12 concentration from healthy mouse is around 1ng/mL from my measurement. For other cytokines, such as IFN-γ and TNF, they cannot be detected. I tried the IL12p40/p70 ELISA, and it gave me similar result. I am wondering whether this concentration of IL-12 is normal.

Daxing, it shouldn´t be any detectable IL12 p70 in serum or plasma in a healthy mouse. If there is, it means there is an active infection -probably viral. IL12 p40 is more abundant in mice, but detection at the ng level sounds a bit too high.
Totally agree, when we measured plasma IL12 p70 in normal mice were almost undetectable concentrations.
Levels for IL-12p40 in the serum of healthy and unchallenged SPF mice in the low ng range are normal (for published results see e.g. PMID: 11313382). Upon challenge you easily induce hundreds of ng of p40. As mentioned by others IL-12p70 should never be detected in healthy mice. The ELISA you’ve used was for p40 and recognizes p40 in all its configurations (p40 monomer, p40 homodimer, p70=p35+p40, IL-23=p19+p40) but the form you detect in the serum of healthy mice is almost exclusively p40 monomer.
