Hi, I'm trying to micropropagate a peach/almond hybrid (hansen536). I took the cutting from juvenile trees and cultured them on MS media supplemented with different combinations of growth hormones (BAP, IBA GA3, IAA). The new growth from the buds is fine in the beginning, but once I transfer them onto a new media they start going downhill. They have very slow growth and the leaves start to turn yellow. Eventually, all the leaves turn yellow and the plants just dry up or lose all the leaves. I wasn't sure if there is a nutrient deficiency or I'm just using the wrong combinations of hormone? Thanks for any advice!
Don't grow them on MS medium. It is totally unsuitable. If you look at the literature you will see that most use Q&L or DKW or some special media for Prunus species. Always start "at the library" and use Google Scholar before you grow a plant.

What are you trying to get? Getting roots? If so, you need a special rooting medium for this special hybrid. Try some medium from published literature first as suggested by Barbara (if any). When we do vegetative propagation of tobacco, we don't add any phytohormone.
Establishment of tissue culture ( or regeneration of new tissues from a woody species) from woody plants or even with juvenile tissues are not easy. Look at the literature to chose Medium type and plant growth combination rather than trying random combinations. I strongly suggest to use generally BA or Kin or adding low amount of an auxin (NAA, IBA or NAA) as plant growth regulators. The general reason for any type of chlorosis is the content of the medium used. Try full, half, quarter and double strength of the medium you used. The explant type (plaigotropic or orttropic), the explanting time, and age (current year shoots or a two year old shoots) are also important factors to establish axenic viable culture. Force the dormant buds to obtain herbeceaus explant form. TRY THESE AND then let me know.

Looks like you are studying the development of these peach x almond (PEAL) hybrid into 'rootstocks' for almond production. In the attached paper (regarding advanced peach-almond hybrid production), one reference (Felipe, 1995) mentions root production from micropropagation. You can check out this paper and look into it. It may contain other citations with micropropagation of peach/almond hybrid.

I have some experience trying to micro-propagate woody plants from shoot apexes and have had the same problems. I would recommend using a more robust (larger) shoot tip. Also when you transfer the plants are you transferring them to bottles or petri-dishes? Once they get going, I would move them to bottles.

Just curious...I know this hybrid is mainly used for 'rootstocks' for almond production due to this rootstock's ability for disease-resistant. But can this hybrid grows fruits and set seeds? What do their fruits look like? and edible?
I recommend you to take shoot tip from shoot which producing in the first culture. Then it culture in MS medium supplemented with BA for shoot multiplication. These images obvious you transfer the shoot with nodal stem (ex plant). The nodal stem is aged tissue. This aged tissue prevent cross the food to the young shoot. Please try to culture shoot tip for shoot multiplication. In this way you can to obtain on survive shoots.

I've never try to grow a whole tree out of the Hansen...I think this hybrid should be able to grow fruits but I'm not sure if the fruits are edible though. If it can get good yield and yummy almond..Then there's no reason to graft it right? That's just my theory :)

Thanks for your recommendation. there are shoots tips which I cut off and transfer them on MS...but they just seemed to melt away...so I was trying to wait till the shoot tips to grow a little bit bigger before I cut them off and transfer them, but still they died. :(

He is the author of the paper I attached for you earlier. And you are right, read on (the message from him)...........
"Thanks for your interest in PEAL hybrids. You asked about the fruit & seed. When one crosses a peach x almond (or almond x peach) the F1 hybrid is generally very fertile, producing many fruit. Unfortunately, the fruit are not very edible,- typically having the dry leathery flesh of an almond and a stony pit containing (usually) a bitter seed. One can self-pollinate a F1 tree to get a true F2 population. In that population you will find wide segregation in many traits. You may find trees that are phenotypically almond, and others peach,- still others with diverse combinations of traits. Some of the fleshy-fruited trees may be somewhat edible,- but probably of lower quality than what one is accustomed to."

Wang , did you check for nutrients like Fe , Zn , since there is a clear cut interveinal chlorosis on either side of mid-rib of infected leaves . Sometimes , there could be overlapping of bacterial disease infection with nutrients.
lu , can you supplement more information about the type of soil and any fertilizer , you applied. Such so called nutrient deficient leaves do not recover , but new leaves will be healthier , once such maladies are remediated.
