I am working on saponin biosynthesis in medicinal plants, and need to measure the expression of the gene to identify where in plant tissue the gene is exactly expressing.
I have selected GAPDH gene which seems to be constantly expressed in plant tissue and especially those who store food material in roots and stem. but still bit confused because the unavailability of sequence of same species origin.
You are initially study the chemical pathway of saponin biosynthesis. Then, you can use A4 strain or MTCC for active growing the roots of the medicinal plant. Then compare the control Vs transform roots. Better is root tissue to study the gene expression .
The major house keeping genes are tRNA-leu, 185 ribosomal RNA and several enzumes i.e acyl-coA binding protein, aldo-keto reductase etc for secondary metabolites
