cotyledons and hypocotyl both can be used for callus. you can use 2,4-D 2mg/l and BA 0.5mg/l can be used for generation of fragile callus. These callus can be used for making cell suspension. Since cell are loosely attached, they can be easily homogenized in MS liguid media for cell suspension. For normal type of callus you can can use 1-1.5mg/l zeatin and IAA 0.1mg/l. These type of callus u can use for transformation. Only Zeatin also induces callus formation.

From tomato plant you can get callus using, leaf segments, saples and inter nodes, however leaf seems to be best
